Techniques of Experimental Plant Biology 1400-225TBER
The aim of the course is to train students in applications of a range of methods used in plant biology research (physiology and biochemistry).
We have selected methods which are necessary and most frequently used in numerous types of experiments, such as protein extraction, ion-exchange chromatography, electrophoresis, molecular filtration, metabolite identification, microscopy. These methods have certain limitations in their use in plant biology and require some preliminary preparation of plant material for analyses. It is necessary to acknowledged with the principles of the methods functions, which will be discussed in details during seminars and in classes.
Selected topics:
Methods of protein extractions and identification
Molecular filtration of proteins
Ion-exchange chromatography of proteins
Selective identifications of metabolites by enzymatic methods
Electrophoresis of proteins on polyacrylamide gels, detection methods
Application of HPLC and FPLC in lab practice
Main fields of studies for MISMaP
biotechnology
environmental protection

Learning outcomes
KNOWLEDGE: The graduate knows and understands
• complex research problems in the natural sciences that require in-depth knowledge in the field of exact sciences (K_W02 Bi)
• specialized terminology in the fields of biochemistry, microbiology, mycology, botany, zoology, genetics, molecular biology, and environmental biology, as well as subject-specific literature in these areas (K_W06 Bi)
• in an advanced manner, a variety of research techniques and tools used in the biological sciences, and appropriately plans their application to solve specific tasks (K_W10 Bi)
SKILLS: The graduate is able to
• apply research techniques and tools appropriate to problems in the studied specialization of biological sciences (K_U01 Bi)
• demonstrate the ability to critically analyze and select biological information, especially from electronic sources and media (K_U04 Bi)
• identify probable causes of experimental failure using professional literature and appropriately modify the course of the experiment (K_U05 Bi)
• under the supervision of a scientific advisor, plan and conduct an experiment using the methods learned; is able to propose methods for carrying out specified determinations and assess the usefulness and limitations of these methods for the studied material (K_U07 Bi)
• critically present research work in the field of the selected specialization of biological sciences using verbal communication and multimedia (K_U09 Bi)
SOCIAL COMPETENCIES: The graduate is prepared to
• fulfill an educational role in society in activities based on biological knowledge and skills (K_K02 Bi)
• communicate to society the latest achievements in the natural sciences and explain the rationale for conducting fundamental scientific research (K_K03 Bi)
• work as part of a team, conducting their own research and co-organizing the work of the entire group (K_K04 Bi)
• demonstrate responsibility for assessing risks associated with the research techniques used and for creating ergonomic and safe working conditions (K_K08 Bi)
Assessment criteria
No more than single absence is allowed during laboratory exercises. Written exam (open questions, test) in which knowledge presented during the seminars and gained during the laboratory will be evaluate.
