The lecture covers the following topics:
1. Construction of a liquid chromatograph,
2. Retention parameters determined on the basis of chromatograms: retention coefficient k, selectivity coefficient, resolution Rs, efficiency N. Parameters influencing these quantities. Purnell equation,
3. Parameters describing the shape of the peak,
4. Kinetic theory of the chromatographic process,
5. Structure of the stationary phases used in HPLC; types and properties of modified stationary phases,
6. Types of chromatography due to the retention mechanism.
7. Properties of mobile phases used in HPLC,
8. Selection of the type of chromatography with regard to the properties of the sample,
9. Properties of detectors used in HPLC,
10. Solving problems in HPLC
The lecture also covers advanced issues of UPLC and ion chromatography, the following topics:
1. Basics of one and two-column ion chromatography,
2. What is chemical damping,
3. Detectors used in ion chromatography
The lecture will be supplemented with five laboratory exercises:
1. Liquid chromatography. Effect of organic solvent concentration on retention,
2. Liquid chromatography. Effect of pH on retention,
3. Determination of phenolic compounds by HPLC using solid phase extraction (SPE),
4. UPLC (Ultra-efficient / ultra-fast liquid chromatography),
5. Ion chromatography.

Learning outcomes
The student knows and understands what is the process of separation of compounds using HPLC. The student is able to calculate the parameters of the separation. The student is able to choose the separation conditions for mixtures of organic chemical compounds
Assessment criteria
Final exam in the form of a written exam, open questions. The pass mark is to obtain more than 50% of the points.
